Background:Orlistat, a well-established and commonly prescribed weight loss medication, is a lipase inhibitor that is commonly prescribed to patients who are obese and are overweight. The aim of this study was to evaluate the pharmacokinetic properties of orlistat in obese and overweight subjects with obesity. Materials and Methods: We prospectively analyzed the pharmacokinetics of orlistat in obese and overweight subjects with and without obesity using single-dose pharmacokinetic analysis. Results: In obese subjects with obesity, the pharmacokinetic (PK) values of orlistat in the presence of obesity were significantly lower than those of andlistat in the absence of obesity (P<0.01). However, the PK parameters of orlistat in the presence of orlistat were not different from those in the absence of orlistat (P>0.05). Conclusion: Orlistat is well tolerated in obese and overweight subjects. Therefore, the results of this study provide new insights into the pharmacokinetics of orlistat in obese and overweight subjects with obesity.
Orlistat, the active ingredient in Xenical (Xenical®), is commonly prescribed to treat obesity and overweight (BMI, ≥27 kg/m2) and to reduce the risk of heart and kidney disease in obese adults (BMI ≥27 kg/m2) [, ]. Orlistat is available in 120 mg, 120 mg, and 120 mg capsules and is administered intravenously at a daily dose of 1 mg once daily. Although the pharmacokinetic (PK) effects of Orlistat have not been established in obese subjects with obesity (BMI ≥27 kg/m2) [–], it is recommended to use Orlistat at least two hours before or two times daily with meals (1 mg) []. Orlistat has been shown to be safe and well tolerated in obese and overweight subjects with obesity [–]. However, it is not recommended to use Orlistat at the same time as a second dose of Orlistat (i.e., 120 mg or 120 mg once daily) []. Therefore, a single dose of Orlistat at a daily dose of 1 mg once daily is not recommended for patients who have not responded to the treatment. Orlistat is not recommended for patients who have been receiving orlistat for ≥1 year because it has been shown to have adverse effects and is associated with increased risk of death and other serious adverse effects in patients with obesity. Additionally, orlistat has been shown to be well-tolerated in patients with obesity and to be an effective in weight management []. However, orlistat has been found to have a higher rate of side effects [,, ]. A recent meta-analysis of randomized controlled trials (RCT) showed that Orlistat may cause adverse effects in patients with obesity []. Therefore, Orlistat is not recommended for patients with obesity without weight loss, especially in patients with weight-related health problems, because Orlistat has a high rate of adverse effects [].
Orlistat is a lipase inhibitor that is available in 120 mg, 120 mg, and 120 mg capsules and is used for the treatment of patients with overweight and obesity. Orlistat is a lipase inhibitor that is indicated for the treatment of patients with obesity with or without a weight-related health condition. In general, orlistat is administered by intravenous (IV) infusion, with or without food, and a maximum daily dose of 1 mg is administered []. Orlistat is available in both oral and intravenous forms and is administered in a once-daily (continuous) formulation, with or without food, and with a maximum dose of 1 mg []. Orlistat is currently available in 120 mg and 120 mg, 120 mg capsules, and it is administered intravenously at a daily dose of 1 mg []. Orlistat is generally well tolerated in patients with obesity [, ], but it may cause a serious side effect [], including an increased risk of death in patients with cardiovascular disease. The mechanism of Orlistat may be related to its action as an antagonist in the brain and to its interaction with the enzyme that converts andlistat into an active metabolite, which may be beneficial in improving weight loss and preventing the development of type 2 diabetes and other cardiovascular diseases [–]. The mechanism of Orlistat may be related to its interaction with other enzymes, including CYP2D6, CYP2D6B, CYP2D6, and CYP2D6L1, which can be affected by Orlistat []. Furthermore, Orlistat can also be metabolized by CYP3A4, which can inhibit orlistat metabolism in different tissues, including liver [].

Tablets like (the brand name for Orlistat) work by reducing the amount of fat your body absorbs. Ordinarily, fats are broken down during the digestion process by enzymes called lipases. Xenical prevents these enzymes from working effectively, limiting how much fat your body is able to absorb. The remaining, undigested fat is then excreted.
In contrast to Xenical, weight loss injections (or pens) work by suppressing your appetite - particularly your appetite for fatty foods. Some weight loss injections also slow the digestion process down, making you feel fuller for longer. They’re simple to use - check out our for more information.
